حديثنا اليوم عن الWestern blot 😎🥼
قبل ما ندخل في تفاصيل الWestern، في تقنية الblotting عندنا ثلاثة انواع مخصصة لنقل DNA, RNA, and proteins:
1️⃣ Southern = DNA
2️⃣ Northern = RNA
3️⃣ Western = Proteins

لو عندك خليط معقد من البروتينات وتبغى تحدد بروتين معين "مثلا من عينة دم"🤔
أو تبغى تقيس كمية تعبير بروتين معين protein expression 🤔

خطوات الشغل على التكنيك 🧪🥼:

أول خطوة هي تحضير العينة، نستخرج منها البروتين عن طريق mechanical or chemical lysis and normalization (SDS charge proteins with (-ve) charge عشان يسهل فصلها )
ثاني خطوة gel electrophoresis
ونستخدم SDS-PAGE 👌

خطوة الblotting
بعد فصل البروتينات يتم نقل الجل على solid support (مثل Nitrocellulose or polyvinylidene fluoride membrane) ويكون أسفل وأعلى الجل والغشاء filter papers and sponge مثل الموضح في الصورة

جهزنا الطبقات نحتاج electric field لعملية نقل البروتين من الجل إلى الغشاء، ومهم يكون الغشاء في اتجاه الpositive electrode 👌
بعدها خطوة الblocking عشان نضمن عدم ارتباط أي Ab في الغشاء ما نحتاجه.
من الbloocking agents المستخدمة:
Casein or Bovine serum albumin

بعدها يكون عندنا incubation للغشاء مع أول
Ab (non-labelled primary Ab)
ويكون مخصصة للبروتين تبعنا👌 عشان يتم ارتباطه ويكون لنا Ab-Ag complex.
بعدها خطوة الغسل ليتم إزالة أي Ab ما أرتبط.

مثل ما عملنا في الخطوة السابقة incubation للغشاء مع labelled secondary Ab وميزته أنه Ab aganist primary Ab (anti-antibody) عشان يقدر يرتبط مع الجسم المضاد الأولي ويكون لنا الcomplex

مرتبط بإنزايم HRP نحتاج للsubstrate ليعطينا visible colored product ويظهر لنا كband على الغشاء👌

Types of detection:
1️⃣-Fluorescent detection (secondary Ab labeled with fluorescent dayes

2️⃣-Chemiluminescent and colorimetric detection (secondary Ab labeled with enzyme reporters, signal generating substrate are used).

وهنا نستخدم radioactive labels بدل الانزايم وما نحتاج بالطبع للsubstrate

Analysis:
-The molecular weight of the protein can be estimated by comparison with marker proteins.
-The amount of proteins can be determined as this is related to band intensity.
